What is BPC-157?

BPC 157 is a synthetic, stable pentadecapeptide (sequence: Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro Ala-Asp-Asp-Ala-Gly-Leu-Val) originally derived from a protective protein found in human gastric juice. It exhibits remarkable physicochemical properties, including resistance to enzymatic degradation and acidic environments, which are often limiting factors for peptide-based compounds (Józwiak et al. 2025). These characteristics, combined with its demonstrated bioactivity across diverse tissue types, suggest it may serve as a model compound for further pharmacological exploration.

In preclinical models, BPC 157 has been shown to modulate a broad range of cellular and molecular pathways. These include the nitric oxide (NO) signaling system, angiogenic responses via VEGFR2, and antioxidant defense mechanisms involving HO-1 and other key enzymes (Józwiak et al. 2025). It has been observed to promote angiogenesis and stimulate granulation tissue formation, and accelerate extracellular matrix remodeling, particularly in models of skin, muscle, and gastrointestinal injury (Seiwerth et al. 2021; Seiwerth et al. 1997). Quantitative histological analyses have demonstrated increased collagen and reticulin synthesis, enhanced neovascularization, and improved wound tensile strength in rodent models following BPC 157 administration (Seiwerth et al. 1997; Vukojević et al. 2021).

Pharmacokinetic studies in animals suggest that BPC 157 is rapidly absorbed via intramuscular routes, and it is distributed to a range of tissues including liver, kidney, muscle, and intestine. It is metabolized into several peptide fragments, primarily involving proline-rich sequences, and eliminated with a half-life under 30 minutes. Despite low CNS penetration, some studies report detectable levels in the brain, supporting its potential for systemic biological effects without significant accumulation (Józwiak et al. 2025).
